Heavy rainfall events, increasing in frequency and intensity with climate change, impact on the quality of the water resource used for drinking-water production. Small-scale water suppliers are particularly sensitive because of their management and the related difficulties of adapting treatment to variations. Decision-support systems, based on monitoring and analytical tools, need to be developed to improve crisis-management procedures related to such events. After presenting the issues related to heavy rainfall events, the article summarizes the tools currently used for quality control of drinking water within this framework, the need for developments and other requirements.

Carbon dots (CDs) comprise a recently discovered class of strongly fluorescent, emission-color-tuning and non-blinking nanoparticles with great analytical and bioanalytical potential. Raw CDs can be obtained by laser ablation or electrochemical exfoliation of graphite, from soot, or thermal carbonization, acid dehydration or ultrasonic treatment of molecular precursors. Passivation of raw CDs makes them fluorescent and their functionalization confers reactivity towards selected targets. CDs can be excited by single-photon (ultraviolet or near-ultraviolet) and multi-photon (red or near-infrared) excitation, and their luminescence properties are due to surface defects. Since the electrochemiluminescence (ECL) of quantum dots (QDs) of silicon was reported by Science in 2002, lots of QDs (e.g., II-VI, III-V and IV-VI) with different sizes and shapes have been used as ECL emitters for bioanalysis. Especially, QDs functionalized with multitudinous biomolecules offer excellent ECL signal-transduction platforms for designing a new generation of biosensing devices.In this article, we focus on recent advances in the ECL principles of functional QDs, and their bioanalytical applications in DNA analysis, immunoassay, cytosensing and detection of other biological molecules.

We present a critical review of the analytical procedures proposed in the past decade for the determination of fluoroquinolones (FQs) by chromatographic methods in solid environmental matrices. We emphasize the overall analytical procedure, from sampling to final detection. We devote special attention to sample preparation, highlighting the problems involved in the analysis of real complex matrices, mainly soil, sediment, manure and sludge. We compare the different extraction techniques in terms of efficiency, time taken and environmental impact, pointing out advantages and drawbacks. Genevac and Dionex sign worldwide distribution agreement

Genevac Ltd (Ipswich, UK), the solvent evaporation business of SP Industries Inc (Warminster, PA) has entered into an OEM agreement with ion chromatography (IC) specialist Dionex Corp (Sunnyvale, CA), which sees the Genevac Rocket™ high speed evaporator being sold as part of Dionex accelerated solvent extraction (ASE®) instruments.

The Rocket high speed evaporator enables rapid concentration or evaporation of up to six 400ml flasks or up to 18 Dionex ASE® vials simultaneously.  In addition using Genevac's proprietary SampleGenie™ technology in conjunction with the Rocket evaporator enables samples to be concentrated directly into an autosampler or storage vial, eliminating the need for manual sample transfer.  To automate transfers from ASE® vials the new Genevac Flip-Flop™ system can be used.  The elimination of manual transfer steps and the automation of the evaporation / concentration process delivers improve laboratory workflow, increases reproducibility of results and maximises sample recovery.

Automation of a Magnetic Bead Workflow for Protein Biomarker Quantification

Agilent Automation Solutions has released a new application note that demonstrates how its Bravo liquid handling system provides an efficient platform for automation of a SISCAPA Magnetic Bead Workflow for protein biomarker quantification by Mass Spectrometry.

Mass Spectrometry (MS) has become the benchmark technology for analysis of peptides based upon its ability to deliver precise quantification, near absolute structural specificity and multiplexing of measurements in a single analytical run. These advantages can be exploited in the analysis of peptides which occur in a single sequence to reveal the concentration of the parent proteins in digests of complex samples such as patient plasma or serum.

The authors of this new application note describe an automated protocol implementing SISCAPA immunoaffinity enrichment of biomarker peptides prior to quantification by MS. In the described protocol, magnetic beads coated with anti-peptide antibodies were used to bind specific target peptides from tryptic digests of plasma and serum samples, after which the beads were washed to remove unbound peptides, and the bound, purified peptides were eluted in small volumes for injection and analysis in an LC/MS/MS system.  Stable isotope labeled versions of the same peptides was used as internal standards to enable accurate quantification.  The Agilent Bravo automated protocol for this application enabled SISCAPA processing of 96 samples in less than 30 minutes, less than half the time required by traditional manual methods.

Daylight Solutions Grows the Aries™ Line of High-Power,


Daylight Solutions, Inc., a leading manufacturer of advanced molecular detection, spectroscopic, and high-power illumination solutions based on quantum cascade and other technologies, announced today the expansion of the Aries™ Series of highpower, multi-wavelength mid-IR lasers. Earlier this year Daylight Solutions introduced the first in the product line, the Aries™-100 air-cooled 2 W laser. The Aries™ commercial product line has now been expanded to include the Aries™-200, -400, and -700 products. The modularity of the Aries™ family of products can now provide the end user with customizable performance in wavelength and power. Our customers can now choose to populate one, two, four, or seven laser “sockets” with any wavelength and power that Daylight offers. Populating all seven sockets in the Aries™-700 can provide over 10 W of cw, air-cooled mid-IR laser power.

“The flexibility and performance of the Aries™ product family is beyond anything available on the market to date,” says Michael Radunsky, Product Marketing Manager for Daylight Solutions’ scientific products. “These turn-key systems can be configured for high power at a single wavelength or to deliver up to seven different wavelengths simultaneously and collinearly.” The Aries™-100 is CDRH approved and is ready for lab use. The higher models in the series are OEM laser systems that are ready for integration. Wavelengths ranging from 2 to 13 μm are available in the Aries™ products. The higher models in the series are also highly ruggedized. Standard performance includes operation within an ambient temperature range of 15 to 40 °C. However, they can also be specified to operate over a much wider range of operating temperatures, ranging from -40 to +70 °C. Aries™ has also been tested for shake and vibration resistance. Optical fiber delivery is also available as an option. “Releasing an additional three models simultaneously for the Aries™ series is a good accomplishment for our team,” states Paul Larson, President of Daylight Solutions. “The flexibility of these models enable applications in multiple markets.”

Peristaltic Pump Simplifies Challenging Liquid Handling Tasks

INTEGRA has launched a new version of its popular DOSE IT peristaltic pump  with enhanced functionality enabling laboratories to now undertake even challenging liquid handling tasks.

Some liquid handling applications require more than just simple pumping.  The new DOSE IT "CUSTOMIZE" software mode allows users to create their own personalized liquid handling protocols. Up to 20 consecutive dispensing and aspirating steps can be defined. A wide range of settable parameters, such as volume, flow rate and repetitions, can be assigned to each step. In addition, the flow rate can be set to ramp up or down to the target speed, thus avoiding foaming, splashing or unwanted turbulences during dispensing.

New Solid State ChIP Technology for Epigenetics

Porvair Filtration Group has launched a new web microsite (www.chromatrap.com) dedicated to providing detailed information on its ground breaking solid state Chromatin Immunoprecipitation (ChIP) technology for the Epigenetics market. The web microsite provides an informative background as to what ChIP technology is about and introduces the new solid state approach pioneered by researchers at Porvair Filtration Group.

Porvair's new Chromatrap® Pro-A technology depends on a porous inert polymer called BioVyon™. Each Chromatrap® spin column contains a BioVyon™ disc with the internal pore structure covalently bonded with Protein A (BioVyon™ Protein A). Complex mixtures containing the chromatin fragments tagged with the antibody of choice are flowed through the column assisted by centrifugation. The column retains the chromatin of interest whilst unwanted debris is flushed away. Purified chromatin is eluted ready for downstream processing and analysis.

A collaborative case study on the site compares the Chromatrap protocol with results from using a Protein G Magnetic Beads technique. Under optimal conditions, the Chromatrap protocol resulted in a 25.5 fold increase in the percentage DNA pull-down using the exact same sample of MCF7 breast cancer cells, targeting the RNA polymerase II protein bound to the GAPDH target gene.

A further independent case study compares Porvair Chromatrap technology with alternative traditional ChIP methods (Sepharose and Dynabeads). In this study Chromatrap columns were shown to provide a greater level of enrichment of the DNA of interest due to low levels of background binding compared to specific antibody binding. Also the DNA pull down (percentage input) was significantly higher for the Chromatrap columns than for the other matrices in these assays.

Traceable® Digital-Bottle™ Refrigerator/Freezer Thermometer

Control Company’s new Traceable® Digital-Bottle™ Refrigerator/Freezer Thermometer will accurately monitor temperatures in freezers, refrigerators, incubators, and environmental chambers with this temperature-buffered sensor. Bottle and solution insulate sensor from rapid changes, as an example, when a refrigerator door is opened. Solution mimics exact temperature of samples. Stainless-steel sensor/probe assures long product life. Entire unit is waterproof.
Easy-to-read digital display shows high, low, and current temperatures. MIN/MAX monitors high/low readings overnight, on weekends, or for any time period—a significant advantage over impossible to read glass thermometers displaying only the current temperature. The entire unit including bottle and display may be placed in any environment within the operating temperature range of –30 to 50.0°C (–22 to 122°F). The resolution is 0.1°. Accuracy is ±1.0°C.

Oxford Instruments receives Queen’s Award for Enterprise: International Trade

Oxford Instruments Plasma Technology has been awarded The Queen’s Award for Enterprise: International Trade for more than doubling its overseas earnings over six years of sustained growth, and exporting over 90% of its production.
Oxford Instruments Plasma Technology, part of the Oxford Instruments Group, designs and manufactures high performance capital equipment for the semiconductor industry. The company started trading in 1981 and is the world’s largest supplier of thin film plasma etch and deposition equipment to the R&D market, and is also in the top two in several key production applications.
The Company’s outstanding performance was based on a comprehensive product range addressing a wide spread of markets combined with an aggressive strategy of developing new markets, primarily in Asia. CHN Microanalysis: A Key Technique for Sample Characterisation

A white paper written by Exeter Analytical describes how CHN microanalysis provides a powerful and straightforward method for determining whether a sample is pure - a key aspect of sample characterisation.

The paper discuss how by providing a precise and accurate analysis of the percentage Carbon, Hydrogen and Nitrogen in an organic sample - CHN microanalysis can compliment techniques such as Mass Spectrometry (MS) and Nuclear Magnetic Resonance (NMR) Spectrometry, thus enhancing the reliability of your analytical results.

Many organisations today benefit from the relative ease with which NMR can be used to determine target compound validity or elucidate the structure of an unknown in the absence of crystal data.  Similarly Mass spectrometry is a powerful analytical technique used by many laboratories to identify unknown compounds, quantify known materials, and elucidate the molecular structure and chemical composition of organic and inorganic substances.

However both NMR and MS techniques have limitations when it comes to assessing sample purity.  If the impurity in the sample is inorganic (e.g. silica from sample purification by flash chromatography using an acidified mobile phase) then it will be invisible to traditional 1H, 13C or 15N NMR techniques.  Likewise organic contaminants present at less than c. 1% of the sample are typically invisible to NMR.   Data is provided showing how CHN analysis can help determine low levels of impurities. Further data is provided showing how CHN microanalysis can be used to determine the presence of residual solvent in samples at levels that has little or no effect on the Mass or NMR spectra.

Agilent Announces Automation Meeting

Titrator Can Run Four Stations in Parallel!

JMScience’s  new  Potentiometric Titrator (COM-1700) allows up to four different titrations to run in parallel at the same time! The COM-1700 has reliable high-speed communications with no response time lag and results appear in real-time. The compact design reduces bench space by 25%. The new buret design allows automatic reagent exchange quickly and efficiently. Longer-life syringe with top dead-center rest position with minimized dead-space and buret head position sensor eliminates improper assembly. The new sample changer accommodates a wide selection of test tubes, beakers and conical flasks. It also is equipped with an auto shut-down function.

Large sized color touch screen allows for easy monitoring of any station and provides for real-time pH calibration and stability. One may easily store data on a USB jump drive. Reading, recalculation and redetection of titration results, and use of data by using other application software is possible by simply downloading the data to a laptop or PC for storage or processing. Data may then be exported to any of several types of file formats including Excel, CSV, HTML, etc. The New COM-1700 also has a built-in thermal printer with easy paper-loading features.

Optimised Filter Plates

UV Clear Bottomed Microplates

Available from Porvair Sciences Ltd - the Krystal UV range of high specification, quartz-bottomed microplates are designed for scientists using assay chemistries which require excitation or detection in the far UV spectral region. 

Precision engineered and assembled - Krystal UV plates provide consistently low background and excellent photometric accuracy down to 220nm.  Manufactured from ultra-pure grade black polystyrene - the plates exhibit very low auto fluorescence and are resistant to solvents including alcohols, DMSO and PBS. Hoefer Inc. has announced the introduction of the NEW Improved MacroVue™ UV-25 Variable Intensity Transilluminator.
The NEW MacroVue™ UV-25 Variable Intensity Transilluminator can now be adjusted from  0 to 9,000µW/cm²  covering a  larger range of preparartive and analytical applications.
The six 8 Watt 302 nm lamps insure uniform illumination of the 21 x 26cm viewing area.  A hinged clear UV safety cover minimizes personnel exposure to UV rays from the lamps.

A NEW Generation of SpinColumns™

SpinColumns™, by Harvard Apparatus, are ideal for quickly desalting, removing unbound markers, and for purification of Proteins, Peptides and DNA.
Harvard Apparatus’ NEW Generation of SpinColumns™ are designed for purification of small samples (10µl to 150µl) in either single column or high-throughput 96-well or 384-well formats.  Centrifugal force drives the separation process in the SpinColumns™.
SpinColumns™ are offered pre-filled with a wide selection of chromatographic materials including gel-filtration, ion-exchange, silica-based reverse (C18) and normal phase materials.  Specific materials such as charcoal, cellulose, and IMAC contained by frits and/or caps are also available.

PBA 2011 - Early bird registration

We would like to inform that the 23rd International Symposium on Pharmaceutical and Biomedical Analysis (PBA 2011 Brazil) early bird registration is now open.

All abstracts presented at the symposium (either oral or poster presentations) are eligible to be submitted as a full paper to the special issue of the Journal of Pharmaceutical and Biomedical Analysis, which will be peer-reviewed following its conventional rules. Please notice that it would be allowed two abstracts per registration.
